目的:用分子标记技术对中药溪黄草的基原植物分类鉴定。方法:用离心柱法提取5种溪黄草基原植物基因组DNA,用85个10 bp的寡核苷酸随机引物进行PCR(聚合酶链式反应),将扩增产物进行琼脂糖凝胶电泳,分析电泳图谱。结果:从85条随机引物中筛选出了12条多态性较好的引物,其中7个引物的扩增图谱表现较好的多态性,且重复性较好。结论:RAPD技术可以成功鉴定溪黄草药材基原。
Objective: To classify and identify the primordial plants of Chinese medicine Xihuangcao by molecular marker technique. Methods: Genomic DNA of five species of Gentiana officinalis was extracted by centrifugal column method. PCR was carried out using 85 oligonucleotide primers of 10 bp in length. The PCR products were analyzed by agarose gel electrophoresis Electrophoresis. Results: Twelve primers with good polymorphism were screened from 85 random primers. Among them, 7 primers showed better polymorphism and better reproducibility. Conclusion: RAPD technique can successfully identify Xihuangcao medicinal material base.
