采用RT-PCR和RACE方法获得了东亚飞蝗(Locusta migratoria manilensis)Toll-9受体基因(LmToll-9)的部分cDNA序列(GeneBank登录号:EU573213),分析了该基因的组织表达特征.获得的cDNA包括3端,长1 231 bp,含一个长918 bp的开放阅读框,编码305个氨基酸,推测的氨基酸序列与其它昆虫的Toll-9受体基因有较高的相似性.东亚飞蝗Toll-9受体具有昆虫Toll受体家族的典型结构,包括胞内的TIR结构域和跨膜区域.半定量RT-PCR研究表明,LmToll-9基因只在东亚飞蝗的中肠组织中表达,而在东亚飞蝗的头部、脂肪体、后腿和血细胞中均没有发现LmToll-9基因的转录.
The partial cDNA sequence of Toll-9 receptor gene (Locusta migratoria manilensis) gene LmToll-9 (GeneBank accession number: EU573213) was obtained by RT-PCR and RACE, and the tissue expression characteristics of the gene were analyzed. The cDNA contains 3 open reading frames (1 231 bp) and a 918 bp open reading frame (ORF) encoding 305 amino acids.The deduced amino acid sequence has high similarity with the Toll-9 receptor gene of other insects. Toll-9 receptor has the typical structure of the insect Toll receptor family, including intracellular TIR domain and transmembrane region.Semi-quantitative RT-PCR studies showed that LmToll-9 gene was only expressed in midgut tissues of oriental migratory locust However, no transcription of LmToll-9 gene was found in the head, fat body, hind leg and blood cells of the migratory locusts.
