用Fenton反应产生羟基自由基(OH·),用电子自旋捕集技术(EPR)测定了四甲基哌啶氮氧自由基及衍生物(4-TEMPO)对OH·的捕获能力。用吩嗪硫酸二甲酯(PMS)-NADH体系产生超氧阴离子自由基(O—·2),用NBT还原法比较了4-TEMPO对O—·2的清除能力。结果显示:①两个四甲基羟哌啶(HTMPOH、OTMPOH)和两个四甲基哌啶氮氧自由基(HTMPO、OTMPO)在40~160μmol·L-1能清除O—·2。②4-TEMPO在抗氧化浓度甚至低于此浓度均使DMPO-OH·(二甲基吡咯啉氮氧化合物与OH·形成的加合物)的信号明显高于对照。两个阴性对照四甲基哌啶(HTMP、OTMP)对O—·2·没有清除作用
Hydroxyl radical (OH ·) was generated by Fenton reaction, and the capture ability of tetramethylpiperidine nitroxide and its derivative (4-TEMPO) on OH · was determined by electron spin-trapping technique (EPR). The superoxide anion radical (O- · 2) was produced by the phenazine methosulfate (PMS) -NADH system and the scavenging ability of 4-TEMPO to O- · 2 was compared by NBT reduction method. The results showed that: ① HTMPO (OTMPOH) and two OTMPO (OTMPO) scavenging O-2 at 40 ~ 160μmol·L -1. ② The signal of DMPO-OH · (adducts formed by dimethylpyrroline oxynitride and OH ·) of 4-TEMPO even at or below the anti-oxidant concentration was significantly higher than that of the control. Two negative control tetramethylpiperidine (HTMP, OTMP) had no scavenging effect on 0-2
