目的:观察通络止痛方对人膝骨关节炎滑膜炎症细胞上清白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)含量的影响。方法:取本院行全膝关节置换术患者术中废弃滑膜组织,经I型胶原酶消化后进行细胞培养,取上清液,加入不同浓度通络止痛方干预,分为:(1)高浓度组:2.5 mg/m L;(2)中浓度组:0.25 mg/m L;(3)低浓度组:0.025 mg/m L;(4)对照组:0 mg/m L。应用酶联免疫吸附法检测观察各浓度组对上清液干预后IL-1β及TNF-α的浓度。结果:高中低浓度组较对照组上清液中IL-1β及TNF-α含量明显降低(P<0.05),且下降的幅度与通络止痛方的浓度呈正相关(IL-1β:r=0.38,P<0.05;TNF-α:r=0.41,P<0.05)。IL-1β及TNF-α的高浓度组达到抑制高峰,与中、低浓度组比较,差异亦有统计学意义(P<0.05)。结论:通络止痛方对人膝骨关节炎滑膜炎症细胞上清IL-1β及TNF-α有抑制作用,随浓度增高,抑制程度越明显。
Objective: To observe the effects of Tonglu Zhitong Prescription on the levels of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) in human knee osteoarthritis synovial inflammatory cells. Methods: The patients with total knee arthroplasty removed intraoperative synovial tissue, the type I collagenase digestion after cell culture, the supernatant, adding different concentrations of Tongluzhitongfang intervention, divided into: (1) High concentration group: 2.5 mg / m L; (2) Medium concentration group: 0.25 mg / m L; (3) Low concentration group: 0.025 mg / m L; The concentration of IL-1β and TNF-α in each concentration group after intervention of supernatant was observed by enzyme-linked immunosorbent assay. Results: The levels of IL-1β and TNF-α in supernatant of high school and low concentration group were significantly lower than those in control group (P <0.05), and the decreasing amplitude was positively correlated with the concentration of Tongluzhitongfang (IL-1β: r = 0.38 , P <0.05; TNF-α: r = 0.41, P <0.05). The high concentration of IL-1β and TNF-α reached the peak inhibition, compared with the medium and low concentration group, the difference was also statistically significant (P <0.05). Conclusion: Tonglu Zhitong Prescription can inhibit IL-1β and TNF-α in supernatant of human knee osteoarthritis synovitis cells. With the increase of concentration, the inhibition degree is more obvious.
