目的:探讨肝细胞癌(HCC)组织中NF-kBp65,VEGF和MMP-9的表达及其临床意义。方法:应用免疫组织化学(SP法)检测35例HCC组织,15例癌旁肝组织及10例正常肝组织中NF-kBp65,VEGF和MMP-9蛋白的表达并分析其与临床病理因素的关系。结果:NF-kBp65,VEGF和MMP-9蛋白在HCC组织中高表达率(分别为80%,74%,71%)高于对照组(P<0.01)。NF-kBp65的表达与HCC的Edmondson分级有关,即在Edmondson分级Ⅲ-Ⅳ的癌细胞中NF-kBp65的表达强于Edmondson分级Ⅰ-Ⅱ的(P<0.05);有静脉浸润的HCC组织中NF-kBp65,VEGF和MMP-9的表达高于无静脉浸润者,差异有统计学意义(P<0.05)。但NF-kBp65的表达与年龄,性别,肿瘤大小,AFP水平无关(P>0.05);VEGF和MMP-9的表达与年龄,性别,肿瘤大小,AFP水平,HCC的Edmondson分级无关(P>0.05)。NF-kBp65的表达与VEGF和MMP-9呈正相关(rs1=0.579,P1<0.05;rs2=0.406,P2<0.05)。结论:NF-kBp65,VEGF和MMP-9过表达可能共同参与HCC的演进,联合检测NF-kBp65,VEGF和MMP-9可作为判断HCC恶性程度和预后的指标。
Objective: To investigate the expression of NF-kBp65, VEGF and MMP-9 in hepatocellular carcinoma (HCC) and its clinical significance. Methods: The expressions of NF-kBp65, VEGF and MMP-9 in 35 HCC tissues, 15 adjacent non-cancerous liver tissues and 10 normal liver tissues were detected by immunohistochemistry (SP method) and their relationship with clinicopathological parameters were analyzed . Results: The high expression rates of NF-kBp65, VEGF and MMP-9 in HCC tissues were 80%, 74% and 71% respectively (P <0.01). The expression of NF-kBp65 was related to the Edmondson classification of HCC. The expression of NF-kBp65 in Edmondson grade Ⅲ-Ⅳ cancer cells was stronger than that of Edmondson grade Ⅰ-Ⅱ (P <0.05). The NF The expression of -kBp65, VEGF and MMP-9 were higher than those without venous invasion, the difference was statistically significant (P <0.05). However, the expression of NF-kBp65 was not related to the age, gender, tumor size and AFP (P> 0.05). The expression of VEGF and MMP-9 was not related to the age, sex, tumor size, AFP level and Edmondson grade of HCC ). The expression of NF-kBp65 was positively correlated with VEGF and MMP-9 (rs1 = 0.579, P1 <0.05; rs2 = 0.406, P2 <0.05). Conclusion: The overexpression of NF-kBp65, VEGF and MMP-9 may participate in the development of HCC. Combined detection of NF-κBp65, VEGF and MMP-9 may be used as an indicator of malignancy and prognosis of HCC.
