目的:建立反相高效液相色谱紫外检测测定血清中异丙酚浓度的方法,并观察病人手术血清中异丙酚浓度与意识水平的关系。方法:用乙腈高氯酸(2∶1)沉淀血清蛋白后取上清液,以邻苯二甲酸二丁酯作为内标定量。结果:本法测定的线性范围为0.5μg·ml-1~4.0μg·ml-1,最低检测限为0.1μg·ml-1。血药浓度的测定说明:异丙酚无蓄积作用,维持意识消失的血药浓度为2.1μg·ml-1以上,术后苏醒时的血药浓度约为1.0μg·ml-1。结论:本法简单、快速,适用于手术过程中异丙酚药动学的监测。
Objective: To establish a method for the determination of propofol in serum by reversed - phase high performance liquid chromatography - ultraviolet detection and to observe the relationship between the concentration of propofol and the level of consciousness in surgical serum of patients. Methods: Serum protein was precipitated with acetonitrile-perchloric acid (2: 1), then the supernatant was taken and quantified with dibutyl phthalate as internal standard. Results: The linear range of this method was 0.5μg · ml-1 ~ 4.0μg · ml-1, the lowest detection limit was 0.1μg · ml-1. The determination of plasma concentration showed that propofol did not accumulate, the plasma concentration of 2.1μg · ml-1 to maintain consciousness disappeared, and the blood concentration after the waking was about 1.0μg · ml-1. Conclusion: This method is simple, rapid and suitable for the monitoring of propofol pharmacokinetics during surgery.
