将乙肝病毒(HBV)的反义或正义寡核苷酸,加入能复制HBV的肝癌细胞模型HepG2.2.15中,通过检测其上清中HBsAg和HBeAg的含量,初步分析了反义寡核苷酸潜在的抗HBV能力。针对C基因区1974─1955的反义链(CSP)在15μmol/L浓度下可抑制84.8%的HBsAg表达和40.0%的HBeAg表达,停药2日时抑制率仍分别为93.5%和39.0%。实验期间未发现反义链对上述细胞形态或细胞生长状况有明显影响。结果提示反义寡核苷酸CSP在实验条件下具有一定的抑制细胞HBV表达的能力。
Antisense or sense oligonucleotide of Hepatitis B virus (HBV) was added into HepG2.2.15 hepatoma cell model which can replicate HBV.
