目的:测定不同采收时期野老鹳草中活性成分没食子酸及鞣花酸的含量,为最佳采收期的确定提供依据。方法:采用HPLC对不同采收期的野老鹳草中没食子酸及鞣花酸的含量进行测定。色谱条件:色谱柱为Dia-monsil C18色谱柱(150 mm×4.6 mm,5μm);流动相为乙腈-0.1%磷酸溶液梯度洗脱;流速为1.0 mL/min;检测波长为274 nm;柱温为25℃。结果:没食子酸及鞣花酸的线性范围分别是:0.075~5.00μg(r=0.9995)、0.05~2.00μg(r=0.9995),平均回收率分别是99.88%(RSD=1.19%)、99.08%(RSD=2.81%)。结论:野老鹳草中没食子酸及鞣花酸的含量均在开花期开始上升,于始果期达到最高。
OBJECTIVE: To determine the content of gallic acid and ellagic acid in the geraniums of different times, to provide the basis for the determination of the best harvesting time. Methods: The contents of gallic acid and ellagic acid in the wild geraniums of different harvesting times were determined by HPLC. Chromatographic conditions: Dia-monsil C18 column (150 mm × 4.6 mm, 5 μm); gradient elution with acetonitrile-0.1% phosphoric acid solution; flow rate of 1.0 mL / min; detection wavelength of 274 nm; 25 ° C. Results: The linear ranges of gallic acid and ellagic acid were 0.075 ~ 5.00μg (r = 0.9995) and 0.05 ~ 2.00μg (r = 0.9995), respectively. The average recoveries were 99.88% (RSD = 1.19% (RSD = 2.81%). Conclusion: The content of gallic acid and ellagic acid in Geranium spp. Rose at the flowering stage and reached the peak at the beginning of fruit stage.
