目的基于NF-κB通路探讨薯蓣皂苷含药血清对IL-1β、IL-17和TNF-α诱导大鼠滑膜细胞株RSC-364的影响。方法采用IL-1β、IL-17和TNF-α联合刺激RSC-364建立类风湿性关节炎细胞模型,制备薯蓣皂苷片和雷公藤多苷片含药血清,应用RT-PCR、Western-Blot检测各组中NF-κBP65 mRNA/蛋白、I-κB/蛋白的表达结果。结果与空白组比较,模型组NF-KBP65mRNA/-NR1表达增加,I-κB mRNA/-NR1减少,差异具统计学意义(P<0.05);与模型组比较,中药组、对照组NF-κBp65减少,I-κB mRNA增加,I-κB-NR1蛋白增加,I-κB-NR1蛋白表达明显减少。结论薯蓣皂苷片含药血清可能通过调控I-κB来抑制转录因子NF-κB p65的产生,进而激活NF-κB信号转导通路。
Objective To investigate the effect of diosgenin-containing serum on the synovial cell line RSC-364 induced by IL-1β, IL-17 and TNF-α in rat NF-κB pathway. Methods The rheumatoid arthritis cell model was established by combining IL-1β, IL-17 and TNF-α with RSC-364 to prepare diosgenin tablets and tripterygium glycosides tablets. RT-PCR and Western-Blot The results of NF-κB P65 mRNA / protein, I-κB / protein expression in each group. Results Compared with the blank group, the expression of NF-KBP65 mRNA / -NR1 increased and the level of I-κB mRNA / -NR1 decreased (P <0.05). Compared with the model group, the expressions of NF-κBp65 I-κB mRNA increased, I-κB-NR1 protein increased, I-κB-NR1 protein expression decreased significantly. Conclusion Diosgenin-containing serum may inhibit the production of NF-κB p65 by regulating I-κB, which may activate NF-κB signal transduction pathway.
