目的:研究RhoC对宫颈癌细胞粘附、侵袭和迁移能力的影响。方法:以宫颈癌SiHa细胞株为研究对象,转入RhoC小干扰RNA抑制其表达,通过体外粘附实验、侵袭实验和迁移试验比较SiHa细胞粘附率、侵袭能力和迁移能力的变化。结果:转入RhoC小干扰RNA后,RhoC蛋白的表达明显下调,对细胞外基质的粘附率明显下降(P<0.01),体外侵袭能力和迁移能力明显下降,侵袭抑制率和迁移抑制率分别为(19.94±6.06)%和(19.16±5.26)%。结论:RhoC能明显促进宫颈癌的转移。
Objective: To study the effect of RhoC on the adhesion, invasion and migration of cervical cancer cells. Methods: The SiHa cell line of cervical cancer was selected as the research object and transfected into RhoC small interfering RNA to inhibit its expression. The adhesion rate, invasion ability and migration ability of SiHa cells were compared by in vitro adhesion test, invasion assay and migration assay. Results: The RhoC protein expression was significantly down-regulated and the adhesion rate to extracellular matrix was significantly decreased (P <0.01). The invasion ability and migration ability of RhoC decreased significantly, and the invasion inhibition rate and migration inhibition rate were respectively (19.94 ± 6.06)% and (19.16 ± 5.26)% respectively. Conclusion: RhoC can significantly promote the metastasis of cervical cancer.
