目的探讨变应性鼻炎(allergic rhinitis,AR)患者外周血单个核细胞(peripheral bloodmononuclear cell,PBMC)内两种皮质类固醇(corticosteroids,CS)受体亚型——CS-α和 CS-β表达水平与类固醇治疗效应之间的关系。方法采用半定量 RT-PCR 方法检测类固醇不同效应 AR 患者和健康对照组 PBMC 的 CS-α、CS-β mRNA,采用免疫细胞化学方法检测 CS-α和 CS-β蛋白的表达。结果①类固醇敏感组、类固醇抵抗组及健康人体内均有 CS-α、β mRNA 的表达,但以 CS-α为主,其 CS-α/GAPDH(葡萄糖甘油醛脱氢酶)mRNA 的比值(±s,下同)分别为1.15±0.75,1.63±0.78,1.27±0.51;②类固醇抵抗组 CS-β mRNA 表达明显高于类固醇敏感组和健康对照(P<0.05),3组 CS-β/GAPDH mRNA 的比值分别为1.42±0.73,0.82±0.59,0.80±0.68;③免疫细胞化学结果显示,类固醇抵抗组 PBMC 的 CS-β表达阳性细胞比率明显高于类固醇敏感组及健康对照组(P<0.01),分别为28.8%±9.9%,5.9%±3.2%,5.5%±6.8%。结论变应性鼻炎患者类固醇抵抗可能与 CS-β的表达亢进有关。
Objective To investigate the expression levels of two corticosteroid receptors (CS-α and CS-β) in peripheral blood mononuclear cells (PBMCs) in patients with allergic rhinitis (AR) And steroid treatment effect of the relationship between. Results ① The expression of CS-α and β mRNA in the steroid-sensitive group, steroid-resistant group and healthy volunteers were both CS-α and CS-α / GAPDH (glucose and glyceraldehyde dehydrogenase) ± s, the same below) were 1.15 ± 0.75, 1.63 ± 0.78 and 1.27 ± 0.51, respectively. ② The expression of CS-β mRNA in steroid-resistant group was significantly higher than that in steroid-sensitive group and healthy controls (P <0.05) GAPDH mRNA was 1.42 ± 0.73, 0.82 ± 0.59 and 0.80 ± 0.68, respectively. ③The results of immunocytochemistry showed that the ratio of CS-β positive cells in steroid-resistant PBMC was significantly higher than that in steroid-sensitive group and healthy control group (P < 0.01), respectively, 28.8% ± 9.9%, 5.9% ± 3.2%, 5.5% ± 6.8%. Conclusions Steroid resistance in patients with allergic rhinitis may be related to the increased expression of CS-β.
