目的观察纺锤体检查点蛋白BubR1在人类肝癌组织、正常肝组织以及肝癌细胞系(HepG2)、正常肝细胞系(LO2)有丝分裂期表达水平的差异,以探讨BubR1蛋白在肝癌发生中的作用。方法用荧光定量PCR和Western blot检测BubR1基因在肝癌组织、正常肝组织,以及在2种细胞中用Nocodazole处理前后的mRNA和蛋白的表达水平。结果BubR1基因在正常组织中的表达高于肝癌组织,在Nocodazole处理前2种细胞中表达无显著差异,在处理后2种细胞中表达均增高,但在LO2细胞中上调水平大于HepG2细胞。结论BubR1基因低表达可能在肝癌的发生过程中起重要作用。
Objective To investigate the difference of BubR1 protein expression in human hepatocellular carcinoma, normal liver tissue, HepG2 and LO2 mitosis in order to investigate the role of BubR1 in hepatocarcinogenesis. Methods The mRNA and protein expression of BubR1 gene in hepatocellular carcinoma, normal liver tissue and before and after Nocodazole treatment were detected by real-time PCR and Western blot. Results The expression of BubR1 gene in normal tissues was higher than that in HCC tissues. There was no significant difference in the expression of BubR1 gene between the two kinds of cells before treatment with Nocodazole. The expression of BubR1 gene in two kinds of cells was higher than that in HepG2 cells. Conclusion The low expression of BubR1 gene may play an important role in the carcinogenesis of hepatocellular carcinoma.
