目的:优化微波辅助提取伸筋草总生物碱的工艺条件。方法:采用酸性染料比色法测定总生物碱含量,以石松灵碱为指标成分。在单因素试验基础上,以总生物碱提取率为评价指标,选取液料比、提取温度和提取时间3个因素进行BoxBehnken中心组合设计,采用响应曲面法优化伸筋草总生物碱的微波辅助提取工艺。结果:伸筋草总生物碱的最佳提取工艺为加17倍量甲醇-水-盐酸(85∶15∶1)于72℃提取8 min;总生物碱提取率4.092 mg·g-1,与预测值4.070 mg·g-1的偏差较小。结论:与传统提取工艺相比,优选的微波辅助提取工艺具有时间短、提取率高、能量消耗低等优点。
Objective: To optimize the microwave-assisted extraction of savannah total alkaloids process conditions. Methods: The total alkaloid content was determined by acid dye colorimetric method. Shisongling base was used as an index component. Based on the single factor experiment, taking the total alkaloid extraction rate as the evaluation index, the three factors of ratio of liquid to material, extraction temperature and extraction time were selected for the combination design of BoxBehnken center. The response surface methodology was used to optimize the microwave-assisted Extraction process. Results: The optimal extraction process of total alkaloids from Scylla spp. Was extracted with 17 times amount of methanol-water-hydrochloric acid (85:15:1) at 72 ℃ for 8 min. The extraction rate of total alkaloids was 4.092 mg · g-1, The deviation of the predicted value of 4.070 mg · g-1 is small. Conclusion: Compared with the traditional extraction process, the preferred microwave-assisted extraction process has the advantages of short time, high extraction rate and low energy consumption.
