目的探讨3种阿片受体亚型(MOR、DOR、KOR)在泻剂结肠大鼠模型结肠内的表达变化及意义。方法使用7~8周龄Wistar大鼠20只,体质量(200±20)g,雌雄各半,按随机数字表法分为对照组(n=10)及泻剂结肠组(n=10),单只分笼饲养(环境温度18~28℃,相对湿度40%~80%)。对照组饲以普通饲料,泻剂结肠组饲以含酚酞饲料。通过RT-qPCR及Western blot法对阿片受体在结肠中的表达进行相对定量分析。结果 3种阿片受体亚型在泻剂结肠组及对照组大鼠结肠内均有不同程度的表达。RT-qPCR检测显示泻剂结肠组MOR、DOR及KOR mRNA相对表达水平均明显高于对照组[(5.093±1.410)vs(1.092±0.222),(2.612±0.612)vs(1.032±0.082),(2.607±0.327)vs(0.995±0.109),P<0.01]。Western blot检测显示泻剂结肠组MOR、DOR及KOR的表达较对照组明显升高[(0.361±0.023)vs(0.152±0.021),(0.246±0.059)vs(0.159±0.005),(0.309±0.018)vs(0.201±0.015),P<0.01]。结论各阿片受体亚型在泻剂结肠大鼠结肠中表达均明显增强,提示其对泻剂结肠大鼠结肠运动及分泌功能可能存在明显的抑制作用。
Objective To investigate the expression and significance of three opioid receptor subtypes (MOR, DOR, KOR) in the colon of cathartic colon rat model. Methods 20 Wistar rats aged 7 to 8 weeks were divided into control group (n = 10) and cathartic colon group (n = 10) according to the random number table method. The body weight was (200 ± 20) , Only a separate cage feeding (ambient temperature 18 ~ 28 ℃, relative humidity 40% ~ 80%). Control group fed with normal feed, cathartic colon group fed with phenolphthalein feed. Relative quantitative analysis of opioid receptor expression in the colon was performed by RT-qPCR and Western blot. Results The three opioid receptor subtypes were expressed in different degrees in the colon of the cathartic group and the control group. RT-qPCR showed that the relative expression levels of MOR, DOR and KOR mRNA in laxative colon group were significantly higher than those in control group (5.093 ± 1.410 vs 1.092 ± 0.222, 2.612 ± 0.612 vs 1.032 ± 0.082, 2.607 ± 0.327 vs (0.995 ± 0.109, P <0.01]. Western blot showed that the expressions of MOR, DOR and KOR in the cathartic colon group were significantly higher than those in the control group [(0.361 ± 0.023) vs (0.152 ± 0.021), (0.246 ± 0.059) vs (0.159 ± 0.005), (0.309 ± 0.018 ) vs (0.201 ± 0.015), P <0.01]. Conclusion The expression of each opioid receptor subtype in the colon of the cathartic colon significantly increased, suggesting that it may have a significant inhibitory effect on the colonic motility and secretion of the cathartic colon rats.
