目的:研究商陆生品及不同炮制品的利尿作用,初步探究其利尿作用机制。方法:采用水负荷大鼠模型,将80只SD大鼠随机分为空白组、模型组、商陆皂苷甲(Es A)组、生品组、不同炮制品(醋炙、醋煮、水煮、清蒸)组,每组10只。同时进行正常状态大鼠利尿实验,将70只SD大鼠随机分为空白组,Es A组,生品组及不同炮制品(醋炙、醋煮、水煮、清蒸)组,每组10只。各给药组的剂量分别为0.005,1.8,1.8,1.8,1.8,1.8 g·kg~(-1),通过给予水负荷模型大鼠及正常大鼠不同的药物,分别测定排尿量,并进行水负荷大鼠大鼠睾丸水通道蛋白9(AQP9)因子表达、肾组织AQP2和AQP4因子的表达试验及正常大鼠睾丸AQP9因子的表达试验。结果:综合6 h总尿量,Es A,商陆生品及不同炮制品均对水负荷大鼠表现出利尿作用;对于正常大鼠仅醋煮品表现出利尿作用。聚合酶链式反应(PCR)技术中对于水负荷大鼠,AQP2 mRNA表达均显著降低,AQP4和AQP9 mRNA表达大部分无显著变化;对于正常大鼠,醋煮组AQP9 mRNA表达显著降低,其余各组无显著变化。结论:商陆对于水负荷大鼠表现出利尿作用,但对正常大鼠利尿作用不明显;商陆的利尿作用可能与AQP2和AQP9 mRNA表达下降有关。
OBJECTIVE: To study the diuretic effects of the biogas products and different processed products, and to explore the diuretic mechanism. Methods: Eighty SD rats were randomly divided into blank group, model group, Es A group, raw product group, different processed products (vinegar Sunburn, vinegar, water boiled , Steamed) group, each group of 10. At the same time, normal rats were given diuretic. 70 SD rats were randomly divided into blank group, Es group, raw product group and different processed products (vinegar, vinegar, boiled, steamed) . Results: The total urinary excretion of 6 h, Es A, pokeweed and different processed products all showed diuretic effect on rats with water load. For normal rats, only the vinegar boiled products showed diuretic effect. Polymerase chain reaction (PCR) technology for water stress rats, AQP2 mRNA expression was significantly reduced, AQP4 and AQP9 mRNA expression most of no significant change; for normal rats, vinegar group AQP9 mRNA expression was significantly reduced, the rest of the No significant change in the group. Conclusion: The larvae showed diuretic effect on water-bearing rats, but had no obvious diuretic effect on normal rats. The diuretic effect of lakeside landfill might be related to the decrease of AQP2 and AQP9 mRNA expression.
