目的:比较研究卵巢恶性肿瘤p53基因第5,6,7外显子突变与p53基因蛋白的过度表达和临床生物学行为的相关性.方法:对50例卵巢恶性肿瘤及10例良性肿瘤石蜡切片进行p53蛋白免疫组化检测,其中23例卵巢恶性肿瘤的新鲜标本进行了p53基因第5,6,7外显子PCR-SSCP检测.结果:50例卵巢恶性肿瘤中12例P53蛋白阳性表达24%(12/50).上皮癌29%(9/28)生殖细胞肿瘤10%(1/10)间质细胞瘤25%(2/8),转移瘤25%(1/4)在23例标本PCR-SSCP检测中突变率为35%(8/23)第5,6.7外显子突变率分别为9%(2/23),9%(2/23).17%(4/23).两者均阳性者为2例.即PCR-SSCP检测和蛋白表达均阳性第6.7外显子各1例.结论:①卵巢恶性肿瘤存在p53基因突变蛋白的过度表达.但突变与蛋白阳性表达并非完全一致;②p53基因突变及其蛋白过度表达与卵巢恶性肿瘤组织类型和临床分期无明显相关性
OBJECTIVE: To compare the correlation between mutations in exons 5, 6 and 7 of p53 gene and p53 gene protein overexpression and clinical biological behavior in ovarian cancer. Methods: Fifty cases of ovarian malignant tumors and 10 cases of benign tumor paraffin sections were examined by immunohistochemistry. P53 was detected by PCR-SSCP in exon 5, 6 and 7 of 23 fresh ovarian cancer samples. Results: The positive expression of P53 protein in 12 cases of ovarian malignant tumors was 24% (12/50). Epithelial neoplasia 29% (9/28) Germ cell tumors 10% (1/10) Smell tumor 25% (2/8), metastases 25% (1/4) Mutation in PCR-SSCP detection of 23 specimens The rate of exon 5 and 6 in exon 35% (8/23) was 9% (2/23) and 9% (2/23), respectively. 17% (4/23). Both were positive in 2 cases. That PCR-SSCP detection and protein expression were positive in 6.7 exons in 1 case. Conclusions: ① Overexpression of p53 gene mutation exists in ovarian cancer. However, the mutation was not completely consistent with the protein positive expression. (2) There was no significant correlation between the mutation of p53 gene and its overexpression and the tissue type and clinical stage of ovarian malignant tumor
