目的:研究针刺百会、风池穴对神经性皮炎模型大鼠NF-κB信号通路的影响,探讨针刺百会、风池穴治疗神经性皮炎机制,为临床治疗该病提供有效的针灸处方。方法:采用采用局部皮肤摩擦刺激与慢性轻度不可预见性的应激方法相结合制备神经性皮炎大鼠模型,在造模成功的基础上,给予针刺百会、风池穴,采用ELISA试剂盒、Western blotting方法来检测观察指标。结果:各治疗组大鼠血清中IL-1β、IL-4、IL-6和TNF-α含量明显降低,皮肤中NF-κB p65和COX-2蛋白表达明显降低,IκB蛋白表达明显升高,与模型组相比较,差异具有统计学意义(P<0.01)。结论:针刺百会、风池穴可能通过NF-κB信号通路,调控炎性细胞因子的水平,抑制了神经性皮炎的慢性炎症的级联放大,对神经性皮炎起到治疗作用。
Objective: To study the effects of acupuncture at Baihui and Fengchi acupoints on the NF-κB signal pathway in neurodermatitis model rats and to explore the mechanism of acupuncture at Baihui and Fengchi acupoints for the treatment of neurodermatitis and to provide effective acupuncture for the clinical treatment of this disease prescription. Methods: Neurodermatitis rat model was prepared by combining local skin friction stimulation and chronic mild unpredictable stress method. On the basis of successful modeling, acupuncture at Baihui and Fengchi acupoints was given. ELISA was used Boxes, Western blotting method to detect the indicators. Results: The levels of IL-1β, IL-4, IL-6 and TNF-α in the serum of rats in each treatment group were significantly decreased, the expressions of NF-κB p65 and COX-2 protein in the skin were significantly decreased, Compared with the model group, the difference was statistically significant (P <0.01). Conclusion: Acupuncture at Baihui and Fengchi acupoints may play a therapeutic role in neurodermatitis through NF-κB signaling pathway, regulating the level of inflammatory cytokines, inhibiting cascade enlargement of chronic inflammation of neurodermatitis.
