目的:研究帕米膦酸钠对大鼠正畸牙移动过程中牙周组织压力侧破牙骨质细胞及破骨细胞分化因子(ODF)表达的影响。方法:选择24只6周龄SPF级健康雌性Wistar大鼠,建立正畸牙移动动物模型,每只大鼠上颌分实验侧和对照侧,于安装矫治器前3 d,于实验侧大鼠第一磨牙近中腭侧黏骨膜下注射帕米膦酸钠50μL,对照侧注射0.9%生理盐水50μL,每3 d注射1次。于正畸加力3、7、14 d时分批处死8只大鼠,制作牙周组织切片,观察破牙骨质细胞数量,免疫组化观察ODF的表达情况。采用PASW Statistics 18软件包对实验数据进行统计学处理。结果:实验侧在3、7、14 d时,第一磨牙压力侧破牙骨质细胞的数目均少于对照侧,其中,7、14 d时两侧差异显著(P<0.05);实验侧在3、7、14 d时,第一磨牙压力侧ODF阳性表达均低于对照侧,其中,7、14 d时两侧差异显著(P<0.05)。结论:局部注射二膦酸盐帕米膦酸钠能够减少大鼠正畸牙移动过程中牙周组织压力侧破牙骨质细胞的数量及ODF的阳性表达。
Objective: To study the effect of pamidronate on the expression of odontoblast and osteoclast differentiation factor (ODF) in periodontal tissues during orthodontic tooth movement in rats. Methods: Twenty-four healthy female Wistar rats of 6 weeks old were enrolled in this study. An animal model of orthodontic tooth movement was established. Each rat was divided into two groups: experimental group and control group, A molar in the palatal mucocutaneous submucosal injection of pamidronate 50μL, control side injection of 0.9% saline 50μL, injected once every 3 d. At 3, 7 and 14 days after orthodontic force, 8 rats were sacrificed in batches, and periodontal tissue sections were made. The number of broken alveolar cells was observed, and the expression of ODF was observed by immunohistochemistry. The experimental data were statistically processed using the PASW Statistics 18 software package. Results: On the 3rd, 7th, 14th and 14th day, the numbers of broken bone cement in the pressure side of the first molar were less than those on the control side, with significant difference between the two sides on the 7th and 14th days (P <0.05) At 3, 7 and 14 days, ODF positive expression on the pressure side of the first molar was lower than that on the control side, with significant difference between the two sides on the 7th and 14th days (P <0.05). CONCLUSIONS: Topical injection of bisphosphonates pamidronate can reduce the number of pressure-exposed periodontal tissues and the positive expression of ODF during orthodontic tooth movement in rats.
