应用HPLC-DAD-ESI-IT-TOF-MS n技术对小叶莲甲醇提取物中的主要化学成分进行定性分析。采用Agilent Zorbax SB-C18(4.6 mm×250 mm,5μm)色谱柱,以0.1%甲酸水溶液(A)-乙腈(B)为流动相进行梯度洗脱,流速为1.0 m L.min-1。质谱使用ESI离子源,正、负离子分析模式采集数据,通过高分辨质谱解析,并结合相关文献和对照品数据共指认了54个化合物,包括木脂素类成分18个和黄酮类成分36个,鉴定了其中50个化合物,其中27个化合物为小叶莲中首次报道。HPLC-DAD-ESI-IT-TOF-MS n技术能快速准确地鉴定小叶莲中所含的主要成分,研究结果为控制小叶莲药材质量、阐释其药效物质和作用机制提供了科学依据,同时对进一步开发和利用小叶莲资源具有指导意义。
Qualitative analysis of the main chemical constituents in the methanol extract of Folium lotus with HPLC-DAD-ESI-IT-TOF-MS n was performed. The mobile phase was eluted with 0.1% formic acid in water (A) -acetonitrile (B) using a Agilent Zorbax SB-C18 (4.6 mm × 250 mm, 5 μm) column with a flow rate of 1.0 m L · min-1. Mass spectra were acquired using ESI ion source and positive and negative ion analysis mode, and 54 compounds including 18 compounds of lignans and 36 flavonoids were identified by high-resolution mass spectrometry combined with related literatures and reference data. Among them, 50 compounds were identified, of which 27 compounds were first reported in leaf lotus. The results showed that HPLC-DAD-ESI-IT-TOF-MS n could identify the main components contained in the leaf lotus quickly and accurately. The results provided a scientific basis for controlling the quality of the drug and the mechanism of action. It is instructive to further develop and utilize the resources of Xiaoyu.
