采用双池培养系统研究颗粒细胞(G-C)与内泡膜细胞(T-C)在甾体激素生成中的旁分泌调节作用。实验选用小母猪的卵巢组织,分离出G-C和T-C,巳分别进行单独培养(单培)或双池培养(双培),内地为G-C,外池为T-C。48小时后用放射免疫法(RIA)测定培养液中雌二醇(E2)、睾酮(T)及孕酮(P)含量。结果显示:(1)在单培时G-C产生极少量E2,当其与T-C双培时则分泌大量E2;P和T产量在双培时比单培时均有增加。(2)T-C在单培时产生大量T,当其与G-C双培时,在培液中可测出少量T;P和E2产量在双培时比在单培时均有明显下降。结果表明:在甾体激素生成过程中,G-C和T-C之间呈现一种旁分泌的调节作用。
Dual cell culture system was used to study the paracrine regulation of G-C and T-C in steroidogenesis. In the experiment, g-C and T-C were isolated from gonads of gilts and cultured separately (single culture) or double-culture (double culture) respectively. The inland area was G-C and the outer pool was T-C. After 48 hours, the contents of estradiol (E2), testosterone (T) and progesterone (P) were determined by radioimmunoassay (RIA). The results showed that: (1) G-C produced a very small amount of E2 in single culture and a large amount of E2 when cultured in double culture with T-C; P and T production increased in double culture compared with single culture. (2) When T-C produced a large amount of T in single culture, a small amount of T could be detected in the culture medium when it was double cultured with G-C; the yield of P and E2 was significantly higher in double culture than in single culture decline. The results showed that during the process of steroidogenesis, there is a paracrine regulation between G-C and T-C.
