为了探讨葛根提取物诱导白血病细胞凋亡的作用及机制。以 18例急性非淋巴细胞性白血病 (ANLL)患者骨髓及HL 6 0细胞分别与葛根提取物、高三尖杉酯碱 (HHT)、阿糖胞苷 (Ara -c)及HA(HHT +Ara c)共同培养 2 4h ,应用细胞形态学、DNA凝胶电泳、DNA片段百分率测定等方法观察以上处理对白血病细胞凋亡的影响 ;对其中 10例处理前后的骨髓细胞用免疫组化法进行bcl 2、c myc(B细胞淋巴瘤 /白血病 2、细胞 癌基因家族 )基因蛋白表达检测。结果 :处理各组均见典型凋亡形态和DNA梯状带 ,对照组无 ,葛根提取物处理后的骨髓细胞bcl 2、c myc基因蛋白表达明显下调。结论 :葛根提取物可诱导ANLL细胞和HL 6 0细胞发生凋亡 ;葛根提取物对bcl 2、c myc基因蛋白表达的下调或抑制可能是其促凋亡机制之一。
To explore the effect and mechanism of Kudzu root extract on apoptosis of leukemia cells. 18 cases of acute non-lymphocytic leukemia (ANLL) patients with bone marrow and HL 60 cells and Pueraria lobata extract, homoharringtonine (HHT), cytarabine (Ara-c) and HA (HHT +Ara c) ) Co-cultivation for 24 hours. Cell morphology, DNA gel electrophoresis, and DNA fragment percentage were used to observe the effect of the above treatment on apoptosis of leukemia cells. Bcl 2 was performed on 10 bone marrow cells before and after treatment by immunohistochemistry. , c myc (B cell lymphoma / leukemia 2, cell oncogene family) gene protein expression detection. RESULTS : The typical apoptotic morphology and DNA ladder were observed in all groups. In the control group, the bcl 2 and c myc gene protein expressions of the myeloid cells treated with Kudzu root extract were significantly downregulated. Conclusion :Pueraria lobata extract can induce apoptosis in ANLL cells and HL 60 cells. The reduction or inhibition of bcl 2 and c myc gene protein expression by Pueraria lobata extract may be one of its pro-apoptotic mechanisms.
