建立了两株分泌抗人早幼粒细胞白血病细胞单克隆抗体细胞株,其单克隆抗体效价稳定,高达1:104。以此单抗作载体,高三尖杉酯碱作弹头,用化学方法偶联制成抗小儿白血病生物导弹,用UV谱和CD谱证实偶联成功。体外试验对靶细胞杀伤率达91.70%~97.98%,高于高三尖杉酯碱对靶细胞杀伤率,对人体各种正常细胞无损伤。
Two monoclonal cell lines secreting anti-human promyelocytic leukemia cells were established, and the titer of monoclonal antibody was as high as 1:10. Using this McAb as a carrier, homoharringtonine was used as a warhead, and anti-childhood leukemia bio-missiles were coupled by chemical methods. The coupling was confirmed by UV and CD spectra. In vitro test target cell killing rate of 91.70% ~ 97.98%, higher than homoharringtonine target cell killing rate, the body of a variety of normal cells without damage.
