目的 探讨骨肉瘤细胞凋亡与 p5 3蛋白异常的相关性 ,p5 3基因在骨肉瘤细胞凋亡发生中所起作用及两者与骨肉瘤预后的关系。方法 采用末端脱氧核糖核酸转移酶介导的 Bio- d U TP末端标记技术 (TU NEL)和免疫组化对 40例骨肉瘤活检组织的细胞凋亡指数和 p5 3蛋白表达进行检测。结果 骨肉瘤组织中存在细胞凋亡 (凋亡指数范围为 0 %~ 14% ,平均 3.1%± 3.5 % )和 p5 3蛋白异常表达 (阳性率为 5 5 %、2 2 / 40 )。p5 3蛋白阳性组的凋亡指数明显低于 p5 3蛋白阴性组 (P<0 .0 1)。结论 p5 3异常可能抑制细胞凋亡。细胞凋亡指数、p5 3蛋白表达与骨肉瘤病人的预后有关。
Objective To investigate the correlation between the apoptosis of osteosarcoma cells and the abnormal expression of p53 protein, the role of p5 3 gene in the apoptosis of osteosarcoma cells and the relationship between them and the prognosis of osteosarcoma. Methods The apoptosis index and p5 3 protein expression in 40 osteosarcoma biopsies were detected by Td NEL and TUNEL. Results Apoptosis was found in osteosarcoma (apoptotic index ranged from 0% to 14% with an average of 3.1% ± 3.5%) and abnormal expression of p53 protein (positive rate was 25%, 22/40). The apoptosis index of p5-3 protein positive group was significantly lower than that of p5-3 protein negative group (P <0.01). Conclusion Abnormal p5 3 may inhibit apoptosis. Apoptosis index, p5 3 protein expression and the prognosis of patients with osteosarcoma.
