为探讨青石棉与p53基因突变之间的关系,采用PCR-SSCP分析方法,对8株青石棉诱发的BALB/c3T3转化细胞进行p53基因的突变检测和分析。结果:从8株转化细胞p53基因上的11个外显子中共发现7处突变,其中2个位于外显子4-6,5个位于外显子9-11;大多数检出的异常表现为比野生型多1条带;突变随机地分布于各个青石棉剂量组。由此表明,p53基因突变与体外转化实验中所用的青石棉剂量无关,检出的突变多位于外显子9-11内,与人类肿瘤中常见p53基因突变多发生在外显子5-8的情形有所不同。文内还对青石棉与p53基因突变之间的关系进行了讨论
To investigate the relationship between crocidolite and p53 gene mutation, PCR-SSCP was used to detect and analyze the mutation of p53 gene in 8 crocidolite-induced BALB / c3T3 transformed cells. RESULTS: Seven mutations were found in 11 exons of p53 gene in 8 transformed cells, of which 2 were located in exon 4-6 and 5 were located in exon 9-11. Most of the abnormalities were detected One more band than the wild type; mutations were randomly distributed in each crocidolite dose group. Thus, p53 mutation was not related to the dose of crocidolite used in the in vitro transformation experiments. The mutations detected were mostly located in exon 9-11.
