为了建立一套快速有效的鉴定交粳稻‘常优1号’种子纯度的方法,本研究利用SSR分子标记技术,对三系杂交粳稻‘常优1号’及其父母本的多态性进行了引物筛选和种子纯度鉴定研究。从24对SSR引物中筛选到1对在‘常优1号’和其双亲之间表现为稳定的共显性且能将杂交种及其亲本完全区分开的引物。利用筛选的RM208引物对‘常优1号’200粒种子进行鉴定,结果表明,‘常优1号’种子纯度为96%,与大田1 000株杂交种纯度鉴定结果 98.2%相比,SSR标记与大田种植鉴定结果基本一致。研究结果表明,引物RM208可用于‘常优1号’种子纯度的鉴定,有助于‘常优1号’的推广应用。
In order to establish a rapid and effective method for the identification of the seed purity of cross in Japonica rice “Changyou 1”, SSR markers were used to study the polymorphism of the three-line japonica hybrid rice “Changyou 1” and its parents Primer Screening and Seed Purity Identification. A total of 24 pairs of SSR primers were screened for one pair of primers which showed stable co-dominant between ’Changyou No.1’ and their parents and could completely distinguish the hybrids and their parents. The results showed that the seed purity of ’Changyou 1’ was 96%, compared with 98.2% of the purity identification of 1000 hybrid plants in Daejeon, SSR markers And field planting identification results are basically the same. The results showed that the primer RM208 could be used for the identification of the seed purity of ’Changyou 1’ and was helpful for the popularization and application of ’Changyou 1’.
