目的探讨肺腺癌中环氧化酶2(COX-2)和缺氧诱导因子1α(HIF-1α)表达的相关性。方法通过免疫组化染色检测106例肺腺癌手术标本中COX-2和HIF-1α的表达,应用不同浓度的选择性COX-2抑制剂NS398对在正常和氯化钴模拟缺氧条件下培养的肺腺癌A549细胞进行处理,Westernblot法分别检测各组HIF-1α的表达。结果106例标本中COX-2强阳性表达率41.5%(44/106),弱阳性表达率20.8%(22/106),阴性表达率7.7%(40/106);HIF-1α强阳性表达率34.0%(36/106),弱阳性表达率4.0%(36/106),阴性表达率2.1%(34/106)。COX-2和HIF-1α的表达呈正相关(P<0.01)。常氧条件下HIF-1α表达相对值0.185±0.057,但缺氧条件下则升高至1.011±0.131,而NS-398则可以明显逆转其趋势(P<0.05)。结论COX-2与HIF-1α在肺腺癌中表达正相关,NS-398不仅能够抑制常氧条件下HIF-1α的表达,而且能够剂量依赖性的抑制低氧条件下HIF-1α表达上调。
Objective To investigate the relationship between the expression of cyclooxygenase 2 (COX-2) and hypoxia inducible factor 1α (HIF-1α) in lung adenocarcinoma. Methods The expressions of COX-2 and HIF-1α in 106 cases of lung adenocarcinoma were detected by immunohistochemical staining. The cells were cultured in normal and cobalt chloride simulated hypoxia condition with different concentrations of NS398, a selective COX-2 inhibitor. Of lung adenocarcinoma A549 cells were treated by Western blot method were detected in each group HIF-1α expression. Results The positive rate of COX-2 in 41 cases was 41.5% (44/106), the positive rate was 20.8% (22/106) and the negative rate was 7.7% (40/106) in 106 cases. The strong positive rate of HIF-1α 34.0% (36/106), weakly positive rate was 4.0% (36/106), negative rate was 2.1% (34/106). The expression of COX-2 and HIF-1α were positively correlated (P <0.01). Under normoxic conditions, the relative expression of HIF-1α was 0.185 ± 0.057, but increased to 1.011 ± 0.131 under hypoxia, while NS-398 significantly reversed the trend (P <0.05). Conclusions COX-2 and HIF-1α are positively correlated with the expression of HIF-1α in lung adenocarcinoma. NS-398 can not only inhibit the expression of HIF-1α under normoxia, but also up-regulate the expression of HIF-1α in a dose-dependent manner.
