目的:建立羟基喜树碱微透析探针回收率的测定方法,考察影响回收率的因素,为体内微透析研究提供依据。方法:采用浓度差法(增量法、减量法)测定羟基喜树碱探针回收率,考察流速、浓度、温度对回收率的影响,并考察回收率的稳定性。结果:增量法和减量法测得的回收率一致;回收率与探针周围媒介浓度无关,稳定性良好,随温度升高而升高。结论:微透析技术可用于羟基喜树碱药动学研究,反透析法可用于羟基喜树碱体内微透析回收率的体内测定。
OBJECTIVE: To establish a method for the determination of hydroxycamptothecin microdialysis probe recovery, investigate the factors influencing the recovery, and provide the basis for the study of microdialysis in vivo. Methods: The concentration of hydroxycamptothecin probe was determined by concentration difference method (incremental method and decrement method). The effects of flow rate, concentration and temperature on the recovery were investigated. The stability of the recovery was also investigated. Results: The recoveries were consistent with the incremental and decrement methods. The recoveries were independent of the concentration of the media around the probe. The stability was good and increased with increasing temperature. CONCLUSIONS: Microdialysis can be used for the pharmacokinetics of hydroxycamptothecin, and the reverse dialysis method can be used for the in vivo determination of microtitration recovery of hydroxycamptothecin in vivo.
