目的观察不同浓度黄芩素处理宫颈癌细胞后,细胞外信号调节激酶1/2(ERK1/2)与基质金属蛋白酶2(MMP-2)、MMP-9 mRNA及蛋白水平表达的变化。方法体外培养HeLa细胞株,分为空白对照组、100μmol/L、200μmol/L黄芩素处理组,培养24 h。明胶酶谱法检测上清液中MMP-2、MMP-9的活性变化。反转录PCR法检测ERK1/2及MMP-2、MMP-9的mRNA水平的表达变化。Western blot法检测ERK1/2、p-ERK1/2及MMP-2、MMP-9的蛋白水平的表达变化。结果黄芩素处理24 h后,与空白对照组相比较,MMP-2、MMP-9的活性变化逐渐降低,且呈浓度依赖性(P<0.05)。ERK1/2、p-ERK1/2、MMP-2、MMP-9的mRNA及蛋白表达随着黄芩素浓度的增强逐渐降低(P<0.05)。结论黄芩素通过激活ERK1/2信号通路下调MMP-2、MMP-9的表达。
Objective To observe the changes of ERK1 / 2 and MMP-2, MMP-9 mRNA and protein levels after treatment with different concentrations of baicalein in cervical cancer cells. Methods HeLa cells were cultured in vitro and divided into blank control group, 100μmol / L, 200μmol / L baicalein group and cultured for 24 hours. The activities of MMP-2 and MMP-9 in the supernatant were detected by gelatin zymography. Reverse transcription PCR was used to detect the expression of ERK1 / 2, MMP-2 and MMP-9 mRNA. The protein expression of ERK1 / 2, p-ERK1 / 2, MMP-2 and MMP-9 were detected by Western blot. Results After the baicalein treatment for 24 h, the activity of MMP-2 and MMP-9 decreased gradually in a concentration-dependent manner compared with the blank control group (P <0.05). The mRNA and protein expressions of ERK1 / 2, p-ERK1 / 2, MMP-2 and MMP-9 decreased gradually with the increase of baicalein concentration (P <0.05). Conclusion Baicalein can down-regulate the expression of MMP-2 and MMP-9 by activating the ERK1 / 2 signaling pathway.
