目的研究姜黄素对SKOV-3细胞的抑制作用。方法实验设模型组(加正常生长的SKOV-3细胞)、空白组(加等量培养液)和实验组。实验组又分为7组[5个浓度(5,10,20,40,80μmol·L~(-1))姜黄素组,即C_1-C_5组,顺铂组(10μmol·L~(-1)),20μmol·L~(-1)姜黄素+10μmol·L~(-1)顺铂组(联合组),每组设6个复孔;用四甲基偶氮唑盐微量酶反应比色法(MTT)检测姜黄素对SKOV-3细胞的增殖抑制作用,用Annexin V-FITC/碘化吡啶(PI)双染法检测细胞凋亡情况,PI单染法检测药品对细胞周期的影响,用酶联免疫法测定细胞Bcl-2蛋白表达情况。结果姜黄素对SKOV3抑制作用,呈浓度依赖性和时间依赖性;模型组凋亡率为0.15%,实验-C1、C2、C3、C4、C5组凋亡率分别为3.86%,26.98%,33.76%,47.75%,68.86%,顺铂组凋亡率为53.91%,联合组凋亡率为77.96%。随着姜黄素浓度增加,SKOV-3细胞在G0-G1期细胞数量逐渐增加,G2-M期和S期细胞数量在逐渐减少(P<0.05);姜黄素能抑制SKOV-3细胞Bcl-2蛋白的表达,并呈浓度依赖性。结论姜黄素能抑制SKOV-3的增殖,主要是诱导细胞凋亡率并使细胞周期停滞的共同作用结果。
Objective To study the inhibitory effect of curcumin on SKOV-3 cells. Methods The experimental model group (normal growth SKOV-3 cells), blank group (plus equal volume of culture medium) and experimental group. The experimental group was further divided into seven groups [5 concentrations (5, 10, 20, 40 and 80μmol·L -1) curcumin group, C_1-C_5 group, cisplatin group (10μmol·L -1 ), 20μmol·L -1 curcumin + 10μmol·L -1 cisplatin group (combined group) with 6 replicate wells in each group. MTT assay The inhibition of curcumin on the proliferation of SKOV-3 cells was detected by MTT assay. The apoptosis of SKOV-3 cells was detected by Annexin V-FITC / PI staining. The effect of drugs on cell cycle was detected by PI single staining The protein expression of Bcl-2 was measured by enzyme-linked immunosorbent assay. Results The apoptosis of SKOV3 was inhibited by curcumin in a concentration-dependent and time-dependent manner. The apoptosis rate in model group was 0.15%. The apoptosis rates in groups C1, C2, C3, C4 and C5 were 3.86%, 26.98% and 33.76 %, 47.75%, 68.86% respectively. The apoptosis rate in cisplatin group was 53.91%, and the apoptosis rate in combination group was 77.96%. With the increase of curcumin concentration, the number of SKOV-3 cells in G0-G1 phase increased gradually and the number of G2-M phase and S phase cells decreased (P <0.05). Curcumin inhibited the proliferation of SKOV-3 cells Protein expression, and in a concentration-dependent manner. Conclusion Curcumin can inhibit the proliferation of SKOV-3, which is mainly the result of the induction of apoptosis rate and cell cycle arrest.
