目的 :检验对培养的牛RPE细胞的氧化损伤是否依赖于氧自由基的浓度。方法 :建立牛RPE细胞培养体系 ,利用次黄嘌呤 /黄嘌呤氧化酶 (Hypoxanthine/Xanthineoxidase ,HX/XO)系统产生牛RPE细胞氧化损伤模型。利用MTT法观察氧自由基对培养的RPE细胞增殖的影响。结果 :单独使用HX和XO对RPE细胞生长均无明显影响。当HX和XO联合使用后 ,产生大量的氧自由基 ,牛RPE细胞受到明显的氧化损伤 ,细胞生长被抑制 ,甚至导致细胞死亡 ,并且这种抑制作用对XO有剂量依赖性 ,浓度越高 ,OD值越低。结论 :HX/XO对体外培养的牛RPE细胞的生长有抑制作用。
OBJECTIVE: To test whether oxidative damage to cultured bovine RPE cells is dependent on the concentration of oxygen free radicals. Methods: The bovine RPE cell culture system was established. Hypoxanthine / xanthine oxidase (HX / XO) system was used to generate oxidative damage model of RPE cells. The effect of oxygen free radicals on the proliferation of cultured RPE cells was observed by MTT assay. Results: HX and XO alone had no significant effect on RPE cell growth. When the combination of HX and XO produced a large number of oxygen free radicals, cattle RPE cells were significantly oxidative damage, cell growth was inhibited, and even lead to cell death, and this inhibitory effect on XO dose-dependent, the higher the concentration, The lower the OD value. Conclusion: HX / XO can inhibit the growth of bovine RPE cells cultured in vitro.
