目的探讨人促黄体激素释放激素(luteinizing hormone releasing hormone,LHRH)受体在喉癌组织中的表达及其在喉癌HEp-2细胞膜表面与LHRH-PE40的亲和力。方法取外科手术收集的50份喉鳞癌及20份癌旁组织标本,采用Western blot法检测LHRH受体的表达,免疫荧光分析法检测LHRH受体的亚细胞定位,ELISA法检测HEp-2细胞膜表面LHRH受体与LHRH-PE40的亲和力。结果 LHRH受体在喉癌组织中的表达量明显高于癌旁组织(P<0.001);LHRH受体在喉癌组织中定位于细胞膜和细胞质,而在癌旁组织中定位于整个腺体周边;LHRH受体在HEp-2细胞膜表面与LHRH-PE40的结合显示出线性和饱和性,且可被LHRH直接特异性抑制。结论 LHRH受体在喉癌组织中的过度表达及其在喉癌细胞表面与LHRH-PE40的高亲和力表明,其可能是LHRH-PE40的一个潜在的治疗靶点。
Objective To investigate the expression of luteinizing hormone-releasing hormone (LHRH) receptor in laryngeal carcinoma and its affinity with LHRH-PE40 on the surface of laryngeal carcinoma HEp-2 cell membrane. Methods 50 Laryngeal squamous cell carcinomas and 20 paracancerous tissue specimens collected by surgery were used to detect the expression of LHRH receptor by Western blotting. The subcellular localization of LHRH receptors was detected by immunofluorescence analysis. HEp-2 cell membrane was detected by ELISA. Surface LHRH receptor affinity for LHRH-PE40. Results The expression level of LHRH receptor in laryngeal carcinoma tissue was significantly higher than that in paracancerous tissue (P<0.001). The LHRH receptor was localized in the cell membrane and cytoplasm in laryngeal carcinoma and localized in the para-carcinoma tissue around the entire gland. The binding of LHRH receptor to LHRH-PE40 on the surface of HEp-2 cell membrane showed linearity and saturation, and could be directly specifically inhibited by LHRH. Conclusion The overexpression of LHRH receptor in laryngeal carcinoma and its high affinity with LHRH-PE40 on the surface of laryngeal carcinoma suggest that it may be a potential therapeutic target for LHRH-PE40.
