目的:研究经前舒颗粒对经前期综合征(Premenstrual Syndrome,PMS)肝气郁证大鼠大脑皮层雌激素受体(estrogen receptor,ER)表达水平的影响。方法:束缚造模法复制PMS肝气郁证大鼠模型,旷场实验对模型评价,RT-PCR和Western blot方法分别检测大鼠大脑皮层ERα、ERβ的mRNA及蛋白的表达。结果:经前舒颗粒给药后大鼠旷场实验得分显著升高,大脑皮层中ERα、ERβ的mRNA和蛋白表达均显著降低。结论:大鼠大脑皮层中ER是经前舒颗粒的作用靶点之一,经前舒颗粒可能通过降低大脑皮层中ERα、ERβ的表达而发挥疗效。
Objective: To investigate the effect of Jingqianshu granule on estrogen receptor (ER) expression in cerebral cortex of rats with syndrome of premenstrual syndrome (PMS). METHODS: The model of PMS liver-qi stagnation was established by restraint modeling. The open field test was used to evaluate the model. The mRNA and protein expressions of ERα and ERβ in rat cerebral cortex were detected by RT-PCR and Western blot respectively. Results: The scores of open-field test in rats treated with Jingqianshu tablets were significantly increased, and the mRNA and protein expressions of ERα and ERβ in the cerebral cortex were significantly decreased. Conclusion: The ER in rat cerebral cortex is one of the target of Jingqian Shu granules. Jingqishu granules may play a role in reducing the expression of ERα and ERβ in the cerebral cortex.
