【目的】研究茶多酚主要成分EGCG体外抗甲型流感病毒FM1株感染及抑制病毒复制的作用。【方法】细胞培养测定抗病毒效价,采用细胞病变抑制(CPE)法和空斑减少试验法,在不同试验策略(治疗、保护)下,评价其体外抗流感病毒作用。【结果】CPE法观察及四甲基偶氮唑盐(MTT)法检测结果显示:在治疗及保护模式下,4~32 mg/mL EGCG均有显著抑制病毒、提高细胞存活率的作用(P<0.01)。空斑减少试验结果显示:2种模式下EGCG治疗指数(TI)分别为39.31及43.48,能有效抑制流感病毒,符合CPE法的结果。【结论】EGCG具有一定的体外抗甲型流感病毒FM1株的作用。
【Objective】 To investigate the infection of anti-influenza virus FM1 strain of EGCG, a major component of tea polyphenols, and the inhibition of virus replication. 【Method】 Cell culture was used to determine the antiviral titer. The cytopathic effect (CPE) and plaque reduction assay were used to evaluate the anti-influenza virus activity in vitro under different experimental strategies (treatment and protection). 【Results】 The results of CPE assay and MTT assay showed that 4 ~ 32 mg / mL EGCG significantly inhibited the virus and increased cell viability in both treatment and protection modes (P <0.01). The results of plaque reduction test showed that the therapeutic index (TI) of EGCG in two modes were 39.31 and 43.48, respectively, which could effectively inhibit the influenza virus and conformed to the result of CPE method. 【Conclusion】 EGCG has certain anti-influenza A virus FM1 strain in vitro.
