利用微生物产生的糖苷酶将虎杖中的虎杖苷转化为白藜芦醇,可以提高虎杖中白藜芦醇的含量。本文从虎杖的微生物富集物中分离筛选到一株霉菌XW-2,其发酵制备的糖苷酶粗酶液处理虎杖,能显著提高其白藜芦醇含量。依据形态学特征和18SrDNA序列比对,XW-2菌株鉴定为黑曲霉(Aspergillus niger);产酶培养基的主要成分和pH值经过优化后,发酵制备的糖苷酶粗酶液处理虎杖,虎杖中的白藜芦醇含量达到了9.24 mg/g,是未经处理虎杖中2.27 mg/g的4.1倍。利用黑曲霉发酵制备的糖苷酶粗酶液直接处理虎杖来提高虎杖中白藜芦醇含量,具有方法简单、成本低等优点,在中药现代化中有较高的应用价值。
Using the glucosidase produced by microorganisms, the polydatin in Polygonum cuspidatum can be transformed into resveratrol, which can increase the content of resveratrol in Polygonum cuspidatum. In this paper, a strain of mold fungus XW-2 was isolated from the microbial enrichment of Polygonum cuspidatum, and the crude enzyme of glycosidase prepared by fermentation was used to treat Polygonum cuspidatum, which could significantly increase the content of resveratrol. XW-2 strain was identified as Aspergillus niger according to the morphological characteristics and 18S rDNA sequence alignment. The main components of the enzyme-producing culture medium and pH value were optimized, and the crude enzyme solution prepared by fermentation was treated with Polygonum cuspidatum, Polygonum cuspidatum Resveratrol reached 9.24 mg / g, 4.1 times higher than 2.27 mg / g in the untreated Polygonum cuspidatum. Aspergillus niger fermentation crude glucosidase prepared by direct treatment of Polygonum cuspidatum to improve the content of resveratrol in Polygonum cuspidatum has the advantages of simple method, low cost, in the modernization of Chinese medicine has a higher value.
