目的观察黄连素是否对重症急性胰腺炎(SAP)大鼠模型肠屏障功能具有保护作用。方法将SD大鼠随机分为假手术组(SHAM组),手术未给药组(SAP组)和手术给药组(SAP+RC组),每组12只。SAP组术前5 d每天生理盐水灌胃1次,SAP+RC组术前5 d每天黄连素灌胃1次。手术造模后24 h取材,对大鼠胰腺及小肠进行组织病理学评分,测定大鼠血清中内毒素、二胺氧化酶及胰腺淀粉酶水平。结果 SAP组及SAP+RC组均造模成功。SAP组、SAP+RC组与SHAM组比较,胰腺、小肠组织病理学评分、血清胰淀粉酶、二胺氧化酶、内毒素均显著升高(P<0.05);与SAP组比较,SAP+RC组胰腺组织病理学评分、二胺氧化酶、内毒素均显著降低(P<0.05)。结论黄连素对SAP大鼠模型肠屏障功能具有明确的保护作用。
Objective To observe whether berberine has a protective effect on gut barrier function in rats with severe acute pancreatitis (SAP). Methods SD rats were randomly divided into SHAM group, SAP group and SAP + RC group, 12 in each group. The SAP group received intragastric administration of saline once daily for 5 days and SAP + RC group received berberine once daily for 5 days before operation. Twenty-four hours after operation, the histopathological score of rat pancreas and small intestine was determined. The levels of endotoxin, diamine oxidase and pancreatic amylase in serum of rats were measured. Results Both SAP and SAP + RC groups were successfully established. SAP, SAP + RC group compared with SHAM group, pancreatic and small intestine histopathological score, serum pancreatic amylase, diamine oxidase, endotoxin were significantly increased (P <0.05); compared with SAP group, SAP + RC The histopathological score, diamine oxidase and endotoxin in pancreas were significantly decreased (P <0.05). Conclusion Berberine has a clear protective effect on intestinal barrier function in SAP rat model.
