目的探讨中医平肝熄风 ,凉血泻火治法的抗神经损伤机理。方法建立谷氨酸致体外培养的大鼠海马神经元兴奋毒性损伤模型 ,并用脑溢安含药血清、细胞外调节蛋白激酶 (ERK)阻滞剂PD 980 59进行干预 ,分别检测各组神经元活化的ERK、c jun氨基末端激酶 (JNK)水平及上清液LDH活性。结果脑溢安含药血清能上调谷氨酸损伤后的大鼠海马神经元ERK水平 ,下调JNK水平 ,PD980 59能阻断脑溢安对受损经元的保护作用。结论以平肝熄风、凉血泻火为主要治法的脑溢安含药血清对谷氨酸致培养大鼠海马神经元损伤后的保护作用与MAPK信号转导途径有关 ,脑溢安上调ERK表达 ,下调JNK表达 ,促进细胞生存
Objective To investigate the anti-neuronal damage mechanism of traditional Chinese medicine Pinggan Xifeng, cooling blood and purging fire. Methods The excitotoxicity model of rat hippocampal neurons cultured in vitro with glutamate was established. The rats were treated with Naoyi-An drug-containing serum and extracellular regulatory protein kinase (ERK) blocker PD 980 59. The neurons in each group were detected. Activated ERK, c jun N-terminal kinase (JNK) levels and supernatant LDH activity. Results Naoyi-an drug-containing serum could up-regulate the ERK level of hippocampal neurons after glutamate injury and down-regulate JNK levels. PD980 59 could block the protective effect of Nao Yi An on damaged meridian cells. Conclusion The protective effect of Nao-Yi-an-containing serum with Pinggan Xifeng and Liangxue Xiehe as the main methods for the injury of hippocampal neurons induced by glutamate is related to the MAPK signal transduction pathway.
