目的 :探讨杭白菊提取液对小牛主动脉血管平滑肌细胞 (VSMC)凋亡的影响及可能的作用机制。方法 :取小牛主动脉进行体外 VSMC培养 ,加入杭白菊提取液干预 ,用膜联蛋白 Annexin检测法 ,在流式细胞仪下观察VSMC的凋亡现象 ,同时取上清液在分光光度仪下测定超氧化物歧化酶 (SOD)、丙二醛 (MDA)。结果 :1在流式细胞仪下观察到加入杭白菊提取液后 VSMC凋亡数量减少 ,随着杭白菊提取液浓度的加大 ,细胞凋亡从 (4.4 2 5±0 .6 2 4 ) %降至 (2 .875± 0 .6 4 0 ) % ;2随着杭白菊提取液浓度的增加 ,SOD值由 (1.6 83± 0 .149)× 10 4 U/ L升高到(2 .2 97± 0 .2 30 )× 10 4 U/ L、MDA值由 (16 6 .4 5 4± 5 6 .80 5 ) μmol/ L下降至 (73.0 6 8± 2 7.2 0 3) μmol/ L。结论 :杭白菊提取液具有抑制小牛 VSMC凋亡及抗氧化作用 ,并呈浓度依赖性。
Objective : To investigate the effect of Chrysanthemum morifolium on the apoptosis of calf aortic smooth muscle cells (VSMC) and its possible mechanism. METHODS: The calf aorta was cultured in VSMC in vitro. The Chrysanthemum morifolium extract was added to intervene. Annexinin was used to measure the apoptosis of VSMC under flow cytometry. The supernatant was taken under the spectrophotometer. Superoxide dismutase (SOD) and malondialdehyde (MDA) were measured. RESULTS: 1 The apoptosis of VSMCs was decreased after the addition of Chrysanthemum morifolium extract by flow cytometry. As the concentration of Chrysanthemum morifolium extract increased, the apoptosis rate was (4.4 2 5±0.62) %. It decreased to (2.875±0.64) % ;2 With the increase of the concentration of Chrysanthemum morifolium extract, the SOD value increased from (1.6 83± 0 .149) × 10 4 U/L to (2. 2 At 97± 0 .2 30 )× 10 4 U/L, the MDA value decreased from (16 6 .4 5 4± 5 6 .80 5) μmol/L to (73.0 68 ± 2 7.2 0 3) μmol/L. Conclusion: Chrysanthemum morifolium extract inhibited the apoptosis and antioxidation of calf VSMCs in a concentration-dependent manner.
