目的探讨血浆miR-125a-5p在肝纤维化患者中的表达及意义。方法收集50例健康志愿者及130例慢性乙型肝炎患者的血浆进行miR-125a-5p实时荧光定量聚合酶链反应(real-time PCR)检测。结果相对于正常组,肝纤维化S1-S4组的miR-125a-5p显著增加(P<0.05),且纤维化越严重,miR-125a-5p的表达量就越高(P<0.05)。患者组中肝硬化患者经扶正化瘀等治疗后,治疗后第7 d及第30 d复查miR-125a-5p表达量较治疗前明显减低(P<0.05),且随着治疗的延长,miR-125a-5p的表达表现为持续下降(P<0.05)。相关性分析显示miR-125a-5p表达与child-pugh分级呈现出高度正相关(相关系数r=0.841,P<0.01)。深入研究显示,患者组中HBeAg(+)患者miR-125a-5p表达量明显高于HBeAg(-)患者(P<0.05)。结论血浆miR-125a-5p的测定有助于肝纤维化的诊断及分期,并对肝硬化患者的病情估计及预后治疗有着一定的帮助。
Objective To investigate the expression and significance of plasma miR-125a-5p in patients with liver fibrosis. Methods The plasma of 50 healthy volunteers and 130 patients with chronic hepatitis B were detected by real-time PCR for miR-125a-5p. Results Compared with the normal group, the expression of miR-125a-5p was significantly increased in S1-S4 group (P <0.05), and the more severe the fibrosis was, the higher the expression of miR-125a-5p was (P <0.05). The expression of miR-125a-5p in patients with cirrhosis was significantly lower than that before treatment (P <0.05) after 7 days and 30 days after treatment, and with the prolongation of treatment, miR The expression of -125a-5p showed a continuous decrease (P <0.05). Correlation analysis showed that miR-125a-5p expression was positively correlated with child-pugh classification (r = 0.841, P <0.01). Further studies showed that the expression of miR-125a-5p in patients with HBeAg (+) was significantly higher than that of patients with HBeAg (-) (P <0.05). Conclusion The determination of plasma miR-125a-5p is helpful for the diagnosis and staging of liver fibrosis, and it is helpful to estimate the disease and prognosis of patients with cirrhosis.
