以新疆甜瓜白粉病抗病品种‘PHHL’与感病品种‘XM2’为亲本,构建F2代遗传分离群体,研究了抗病品种‘PHHL’中白粉病抗性基因的遗传规律。采用BSA(Bulked Segregation Analysis)法和SSR(Simple Sequence Repeat)分子标记技术,对白粉病抗性基因进行遗传定位,为抗白粉病育种提供种质资源。结果表明:甜瓜‘PHHL’品系对白粉病Podosphaera xanthii的抗性受一个显性基因控制;筛选11个SSR多态性标记,通过连锁分析,将该基因定位于LGVII标记SSR12510与ECM123之间;327、328号标记发生了偏分离现象且差异显著,这2个标记存在偏分离位点。
A genetic linkage map of F2 progenies was constructed with the resistant cultivar ’PHHL’ and the susceptible cultivar ’XM2’ of Xinjiang melon powdery mildew and the inheritance of powdery mildew resistance genes in resistant cultivar ’PHHL’. Bulbous segregation analysis (BSA) and SSR (Simple Sequence Repeat) molecular markers were used to provide genetic mapping of powdery mildew resistance genes and provide germplasm resources for powdery mildew resistance breeding. The results showed that the resistance of muskmelon ’PHHL’ strain to Podosphaera xanthii was controlled by a dominant gene. Eleven SSR polymorphism markers were screened and the gene was mapped between LGVII marker SSR12510 and ECM123 by linkage analysis. 327 There was a partial segregation phenomenon in the 328th marker, with significant difference. There were partial segregation sites in these two markers.
