目的了解云南和山西2地无偿献血人群丙型肝炎病毒(HCV)的基因型分布及其差异。方法采用荧光定量PCR(Q-PCR),对所收集的203份ELISA检测抗-HCV阳性的无偿献血者标本(云南102份,山西101份)做核酸检测,对核酸阳性的标本做E1基因扩增和测序;E1基因阴性者,进一步做NS5B基因扩增;采用DNASTAR、Bi-oedit和Mega4等软件对E1、NS5B基因进行分析及分子进化研究。结果203份ELISA检测抗-HCV阳性的无偿献血者标本中,云南102份标本经Q-PCR检测出核酸阳性55份,从中扩增出E1基因43份,补做NS5B基因扩增获得阳性结果8份;山西省101份标本经Q-PCR检测出阳性46份,从中扩增出E1基因33份,补做NS5B基因扩增获得阳性结果6份。云南省献血者HCV基因型(亚型)可分为7种:1b[7.84%(4/51)]、2a[13.73%(7/51)]、3a[17.65%(9/51)]、3b[41.18%(21/51)]、6a[7.84%(4/51)]、6n[9.80%(5/51)]和6v[1.96%(1/51)];山西省献血者HCV基因型(亚型)较简单,可分为3种:1b[89.74%(35/39)]、2a[7.69%(3/39)]和3b[2.56%(1/39)]。结论我国云南和山西2地无偿献血人群中的HCV基因型(亚型)的分布存在明显的差异。
Objective To understand the genotype distribution and difference of hepatitis C virus (HCV) in the blood donors in Yunnan and Shanxi. Methods Totally 203 samples of anti-HCV positive blood donors (102 Yunnan and 101 Shanxi) were collected for nucleic acid detection by real-time fluorescence quantitative PCR (Q-PCR), and E1 gene amplification was performed on nucleic acid positive samples Amplification and sequencing; E1 gene negative, further NS5B gene amplification; using DNASTAR, Bi-oedit and Mega4 and other software on E1, NS5B gene analysis and molecular evolution. Results 203 samples of anti-HCV positive anti-HCV positive blood donors were detected in 102 samples of Yunnan by Q-PCR to detect 55 positive nucleic acids, amplified from 43 genes in the E1, NS5B gene complements make a positive result 8 ; Shanxi Province, 101 specimens were detected by Q-PCR, 46 were positive, 33 were amplified from the E1 gene, make NS5B gene amplification obtained 6 positive results. HCV genotypes (subtypes) in Yunnan donors can be divided into seven types: 1b [7.84% (4/51)], 2a [13.73% (7/51)], 3a [17.65% (9/51) 3b (41.18% (21/51)], 6a [7.84% (4/51)], 6n [9.80% (5/51)] and 6v Type (subtype) is relatively simple and can be divided into three types: 1b [89.74% (35/39)], 2a [7.69% (3/39)] and 3b [2.56% (1/39)]. Conclusion There are significant differences in the distribution of HCV genotypes (subtypes) among the blood donors in Yunnan and Shanxi in China.
