目的探讨PPARγ、PTEN、P27kipl三种蛋白在子宫内膜癌中的表达、意义及三者之间的相关性。方法采用免疫组化SP法检测60例子宫内膜癌、35例增生性子宫内膜及15例不典型增生子宫内膜的石蜡标本切片中PPARγ、PTEN、P27kipl蛋白的表达。结果1子宫内膜癌组织与增生性、正常子宫内膜组织比较PPARγ阳性表达率升高(P<0.01),PTEN、P27kipl阳性表达率则降低(P<0.01)。2在子宫内膜癌中,PPARγ的表达率在不同组织学分级、FIGO分期间的差异有统计学意义(P<0.05),在肌层浸润深度、有无淋巴结转移及组织学类型间的差异无统计学意义(P>0.05);PTEN、P27kipl的表达率在组织学分级、FIGO分期、肌层浸润程度间的差异均有统计学意义(P<0.01),而在有无淋巴结转移及组织学类型间差异无统计学意义(P>0.05)。3在子宫内膜癌中,PPARγ与PTEN、PPARγ与P27kipl之间无明显相关性;PTEN与P27kipl表达呈正相关(r=0.636,P<0.01)。结论PPARγ、PTEN、P27kipl与子宫内膜癌的发生和发展有一定关系,它们的表达与常用的预测预后的临床特征有关,因而对它们的检测对肿瘤早期诊断、预后评估有一定价值。
Objective To investigate the expression and significance of PPARγ, PTEN and P27kipl in endometrial carcinoma and their correlations. Methods The expressions of PPARγ, PTEN and P27kipl protein in 60 specimens of endometrial carcinoma, 35 specimens of proliferative endometrium and 15 specimens of atypical hyperplasia were detected by immunohistochemical SP method. Results 1 The expression of PPARγ in endometrial carcinoma was significantly higher than that in proliferative and normal endometrial tissues (P <0.01). The positive expression rates of PTEN and P27kipl were decreased (P <0.01). 2 in endometrial carcinoma, the expression of PPARγ in different histological grade, FIGO staging difference was statistically significant (P <0.05), in the depth of myometrial invasion, lymph node metastasis and histological differences between the types (P> 0.05). The expression rates of PTEN and P27kipl were significantly different between histological grade, FIGO stage and myometrial infiltration (P <0.01), but there was no difference in lymph node metastasis and tissue There was no significant difference between the study types (P> 0.05). 3 There was no significant correlation between PPARγ and PTEN, PPARγ and P27kipl in endometrial carcinoma; PTEN and P27kipl expression were positively correlated (r = 0.636, P <0.01). Conclusion The expression of PPARγ, PTEN and P27kipl is related to the occurrence and development of endometrial carcinoma. Their expression is related to the clinical characteristics of commonly used predictive prognosis. Therefore, their detection has some value in the early diagnosis and prognosis evaluation of tumor.
