目的探讨受体O型蛋白酪氨酸磷酸酶(PTPRO)对妊娠期肝内胆汁淤积症(ICP)患者Th1/Th2细胞偏倚的影响。方法分离获取正常妊娠(A组,30例)、轻型ICP(B组,28例)和重型ICP(C组,30例)孕妇胎盘来源的单核-巨噬细胞,采用RT-PCR和Western blot法分别检测PTPRO mRNA和蛋白的表达,Western blot法检测NF-κB/p65的磷酸化水平。分离获取孕妇外周血T淋巴细胞,与胎盘来源的单核-巨噬细胞共培养,ELISA法检测细胞培养上清液中IFN-γ、IL-12、IL-4和IL-10水平。结果与A组相比,B、C组单核-巨噬细胞PTPRO表达和NF-κB/p65磷酸化水平升高,与其共培养的外周血T淋巴细胞高分泌IFN-γ和IL-12(P<0.05);与B组相比,C组PTPRO表达和NF-κB/p65磷酸化水平升高,IFN-γ和IL-12分泌增多(P<0.05)。结论 ICP患者胎盘单核-巨噬细胞高表达PTPRO,促使T淋巴细胞向Th1细胞偏倚。
Objective To investigate the effect of PTPRO on Th1 / Th2 cell biases in patients with intrahepatic cholestasis of pregnancy (ICP). Methods Placenta-derived mononuclear macrophages from normal pregnant women (group A, 30 cases), mild ICP (group B, 28 cases) and heavy ICP (group C, 30 cases) were isolated and analyzed by RT-PCR and Western blot PTPRO mRNA and protein expression were detected by Western blot, and the phosphorylation of NF-κB / p65 was detected by Western blot. T lymphocytes were isolated from peripheral blood of pregnant women and co-cultured with placenta-derived monocyte-macrophages. The levels of IFN-γ, IL-12, IL-4 and IL-10 in cell culture supernatants were detected by ELISA. Results Compared with group A, PTPRO expression and phosphorylation of NF-κB / p65 in group B and C mononuclear macrophages were increased. Peripheral blood T lymphocytes co-cultured with IFN-γ and IL-12 Compared with group B, PTPRO expression and phosphorylation of NF-κB / p65 increased and secretion of IFN-γ and IL-12 increased in group C (P <0.05). Conclusion ICP patients with placental monocyte - macrophage expression of PTPRO, to promote T lymphocytes to Th1 cell bias.
