目的了解mdm2蛋白的表达与肺腺癌、鳞癌临床病理特征之间的关系及意义,探讨mdm2蛋白在肺癌发生、发展过程中的可能作用。方法用免疫组化法(SP法)检测mdm2蛋白在53例肺癌组织和19例癌旁正常肺组织的表达情况,并结合肺癌的临床病理特征进行对比分析。结果mdm2蛋白定位于细胞质或细胞核,为棕黄色颗粒。53例肺癌组织mdm2蛋白过表达率为52.8% (28/53),19例癌旁正常肺组织均不表达mdm2蛋白,二者差异有显著性(P<0.05)。Ⅲ~Ⅳ期和有淋巴结转移肺癌的mdm2蛋白过表达率分别为71.4%(15/21)和67.7%(21/31),显著高于Ⅰ~Ⅱ期(40.6%)和无淋巴结转移的过表达率(31.8%,P<0.05)。低分化肺癌mdm2蛋白过表达率90.0% (9/10)显著高于高分化及中分化的过表达率(40.0%和45.5%,P<0.05)。mdm2蛋白过表达率与患者年龄、性别、吸烟史及病理类型无显著相关性(P>0.05)。mdm2蛋白过表达组肺癌患者的术后5年生存率显著低于不表达组(P<0.05)。结论mdm2蛋白在肺癌中过表达与淋巴结转移、pTNM分期、分化程度有关。mdm2蛋白过表达可能在肺癌的发生、发展及预后中起重要作用。
Objective To investigate the relationship between the expression of mdm2 protein and the clinicopathological features of lung adenocarcinoma and squamous cell carcinoma and to explore the possible role of mdm2 protein in the occurrence and development of lung cancer. Methods The expression of mdm2 protein in 53 lung cancer tissues and 19 adjacent normal lung tissues was detected by immunohistochemistry (SP method), and compared with the clinicopathological features of lung cancer. Results mdm2 protein located in the cytoplasm or nucleus, as brown particles. The overexpression rates of mdm2 protein in 53 cases of lung cancer were 52.8% (28/53), while no expression of mdm2 protein was detected in 19 cases of normal lung tissues. The difference was significant (P <0.05). The overexpression rates of mdm2 protein in stage Ⅲ ~ Ⅳ and lung cancer with lymph node metastasis were 71.4% (15/21) and 67.7% (21/31) respectively, which were significantly higher than those in stage Ⅰ ~ Ⅱ (40.6%) and without lymph node metastasis The expression rate (31.8%, P <0.05). The overexpression rates of mdm2 protein in poorly differentiated lung cancer were 90.0% (9/10), which were significantly higher than those in well differentiated and moderately differentiated tumors (40.0% and 45.5%, P <0.05). The overexpression rate of mdm2 had no significant correlation with age, sex, smoking history and pathological type (P> 0.05). The 5-year survival rate of lung cancer patients with mdm2 overexpression group was significantly lower than that of non-expression group (P <0.05). Conclusion The overexpression of mdm2 in lung cancer is related to lymph node metastasis, pTNM stage and differentiation degree. Overexpression of mdm2 may play an important role in the occurrence, development and prognosis of lung cancer.
