目的:研究姬松茸多糖对小鼠骨髓来源树突状细胞抗肿瘤功能的影响,为临床应用提供理论依据。方法:体外培养小鼠骨髓来源DC,加入ABP溶液,流式细胞仪检测DC表面CD86和CD11a的表达,混合淋巴细胞反应检测ABP对DC抗原提呈能力的影响,MTT法检测ABP对细胞毒性T淋巴细胞(CTL)对H22肝癌细胞杀伤功能的影响。结果:中剂量ABP组(100μg/mL)DC表面CD86、CD11a表达上调;CTL对肿瘤细胞的杀伤率升高,差异有统计学意义(P<0.05)。结论:适当浓度的ABP能促进DC分化成熟,增强DC的抗肿瘤作用。
Objective: To study the effect of polysaccharides from Agaricus blazei Murill on the anti-tumor function of dendritic cells derived from bone marrow in mice and to provide a theoretical basis for clinical application. Methods: Bone marrow-derived DCs were cultured in vitro. ABCs were added into the bone marrow of mice. The expression of CD86 and CD11a on DCs was detected by flow cytometry. The effect of ABP on the DC antigen presenting ability was detected by mixed lymphocyte reaction. Effect of lymphocyte (CTL) on cytotoxicity of H22 hepatoma cells. Results: The expression of CD86 and CD11a on the surface of DC in medium dose ABP group (100μg / mL) was up-regulated. The cytotoxicity of CTL to tumor cells was increased, the difference was statistically significant (P <0.05). Conclusion: The appropriate concentration of ABP can promote the differentiation and maturation of DC and enhance the anti-tumor effect of DC.
