目的观察分析对衰老大鼠给予何首乌饮后大鼠卵巢组织凋亡相关蛋白FOX、SIRT1、c-Myc表达的影响。方法大鼠给予D-半乳糖连续腹腔注射法制备亚急性衰老大鼠模型,模型制备成功后何首乌饮连续灌胃60d,大鼠断头取卵巢。采用Western Bloting法检测各组大鼠卵巢FOX、SIRT1及c-Myc蛋白表达的变化。结果模型组较正常组卵巢细胞FOX、SIRT1及c-Myc表达量降低;何首乌饮可使卵巢组织FOX、SIRT1及c-Myc表达量增加,并呈现一定的剂量依赖性,以高剂量最佳,差异有统计学意义(P<0.05)。结论何首乌饮方剂能延缓衰老大鼠卵巢细胞凋亡的发生,其机制可能是通过干预凋亡相关基因FOX、SIRT1及c-Myc蛋白表达抑制卵巢细胞的凋亡。
Objective To observe and analyze the effect on the expression of FOX, SIRT1 and c-Myc in rat ovariectomized rats induced by Polygonum multiflorum Thunb. Methods The rat model of subacute aging was established by continuous intraperitoneal injection of D-galactose. After the model was successfully prepared, Polygonum hydropiperum was continuously fed for 60 days, and the rats were decapitated for ovary. Western Bloting was used to detect the expression of FOX, SIRT1 and c-Myc protein in ovary of each group. Results The expression of FOX, SIRT1 and c-Myc in ovary cells of model group decreased significantly. The content of FOX, SIRT1 and c-Myc in ovary tissue of Polygonum multiflorum was increased and the dose of Polygonum multiflorum Thunb. The difference was statistically significant (P <0.05). Conclusion Polygonum multiflorum Decoction can delay the apoptosis of ovarian cells in aging rats. The mechanism may be that the apoptosis of ovarian cells may be inhibited by interfering with the expression of FOX, SIRT1 and c-Myc.
