糖心安对Ⅱ型糖尿病大鼠周围神经病变的保护作用及其机理研究

目的:评价糖心安胶囊对Ⅱ型糖尿病致周围神经病变的防治作用及相关机制。方法:将SD雄性大鼠随机分为两组,即对照组和模型组。所有造模动物灌胃高糖高脂乳,正常对照组给等体积蒸馏水,每天一次,连续4周,然后一次性尾静脉注射链脲菌素25mg/kg以复制2型糖尿病模型。正常对照组动物给予等容积缓冲液。将造模成功大鼠随机分为5组:模型组、盐酸二甲双胍0.1 g/kg+卡托普利0.25g/kg、糖心安0.3 g/kg、糖心安0.15 g/kg、糖心安0.075 g/kg,灌胃给药,模型组和正常对照组分别给于配药溶剂0.5%羧甲基纤维素钠;每周称重,调整给药量,持续给药8周。在末次给药后24小时,测定其坐骨神经运动神经传导速度、血清TG、TC、GLU、SOD、MDA、糖化血清蛋白、AGEs,并取其坐骨神经做病理组织学检测。结果:在造型8周末其模型大鼠血清LDLC含量增加,MDA明显升高,糖化血红蛋白以及糖基化终末产物均显著增加,其坐骨神经传导速度(MNCV)降低,有髓纤维髓鞘空泡变性至严重脱髓鞘,神经纤维变细,部分神经组织局部发生肿胀现象,许旺细胞增殖等病理改变;糖心安0.3g/kg能显著降低血清中MDA、糖化血红蛋白以及AGEs,显著提高坐骨神经传导速度,改善糖尿病大鼠外周神经病理形态学变化。结论:糖心安对Ⅱ型糖尿病周围神经病变具有一定的防治作用,其作用机理可能与其能降低糖尿病大鼠血清糖化血红蛋白、糖基化终末产物以及MDA水平有关,其详细作用机理还有待进一步研究。 Objective: To evaluate the preventive and therapeutic effects of Tang Xin An Capsule on peripheral neuropathy induced by type 2 diabetes mellitus and its related mechanism. Methods: SD male rats were randomly divided into two groups: control group and model group. Results: At the 8th weekend of modeling, serum LDLC level increased, MDA significantly increased, glycated hemoglobin and advanced glycation end products increased significantly, the sciatic nerve conduction velocity (MNCV) decreased, myelin sheath degeneration To severe demyelination, nerve fibers become thinner, part of the nervous tissue swelling occurred locally, Schwann cell proliferation and other pathological changes; Tang Xinan 0.3g / kg can significantly reduce the serum MDA, HbA1c and AGEs, significantly increased sciatic nerve conduction velocity , To improve the pathological changes of peripheral nerve in diabetic rats. CONCLUSION: Tangxin’an has some preventive and therapeutic effects on type 2 diabetic peripheral neuropathy. Its mechanism may be related to the decrease of serum levels of HbA1c, glycosylation end products and MDA in diabetic rats. The mechanism of action is still to be further studied .
