目的观察高糖环境下豚鼠膀胱Cajal间质细胞超微结构的改变,探讨糖尿病膀胱(DCP)的发病机制。方法采用酶消化法原代培养,透射电镜观察不同葡萄糖浓度下培养的ICCs超微结构。结果随糖浓度增高和培养时间延长,ICCs内细胞器明显减少,线粒体大小不等、数量减少、肿胀、空泡样变甚至溶解,内质网扩张、空泡样变,胞质广泛溶解,突起消失。结论高糖环境对豚鼠膀胱ICCs超微结构造成显著影响,这种结构破坏可能是造成DCP发病的重要原因之一。
Objective To observe the ultrastructural changes of Cajal interstitial cells in guinea pig bladder under high glucose and to explore the pathogenesis of diabetic bladder (DCP). Methods Primary culture was performed by enzyme digestion. The ultrastructure of ICCs cultured under different glucose concentrations was observed by transmission electron microscopy. Results With the increase of glucose concentration and culture time, the number of organelles in ICCs was significantly decreased, the number of mitochondria was decreased, the number of mitochondria was decreased, swelling, vacuolization or even dissolution was observed, the endoplasmic reticulum was dilated, the vacuoles were changed, the cytoplasm was widely dissolved and the processes disappeared . Conclusion The high glucose environment has a significant effect on the ultrastructure of bladder ICCs in guinea pigs. This structural disruption may be one of the important causes of DCP.
