石蒜碱内铵或酸盐(AT-1840,Lyc)明显改变DNA的圆二色谱(CD)图。改变的程度与药物浓度有关。Lyc 12.5μg/ml时,DNA的274nm峰位蓝移到266nm,且峰的[ψ]值增加。243nm负峰的[ψ]值增加、谱带变窄,峰位红移到245nm.Lyc的浓度为50μg/ml时,DNA的CD谱完全改变,出现267-289的负峰,207nm负峰及244nm正峰。当浓度达100μg/ml时,溶液出现沉淀,无CD谱。药物与DNA复合物透析后,CD谱可基本恢复到原形,但峰较低。Lyc本身无CD谱。低浓度时,反应约0.33min达平衡,高浓度时,反应约3.96min达平衡。
Lycorisidine or its salts (AT-1840, Lyc) significantly changed the circular dichroism (CD) pattern of DNA. The degree of change is related to the drug concentration. At Lyc 12.5 μg / ml, the peak position of 274 nm of DNA shifted blue to 266 nm, and the peak value of [ψ] increased. The peak of [ψ] at 243nm increased, the band narrowed and the peak shifted to 245nm.When the concentration of Lyc was 50μg / ml, the CD spectrum of DNA changed completely with the negative peak of 267-289 and the negative peak of 207nm 244nm positive peak.
