目的应用膜片钳全细胞记录技术,探讨觉醒肽orexinA对小鼠前额叶皮层PL区细胞的作用。方法制备小鼠前额叶皮质脑片,在膜片钳全细胞记录模式下,观察记录PL区锥体神经元和中间神经元的电生理学特点及其对orex-inA的反应情况。结果54例锥体神经元对orexinA总反应率为51.9%。36例Ih(+)锥体神经元对orexinA反应率为66.7%,18例Ih(-)锥体神经元对orexinA反应率为22.2%,两者差异具有统计学意义(P<0.01);15例中间神经元对orexinA均无反应。结论orexinA对前额叶皮层PL区锥体神经元具有一定兴奋性效应,并且表现该兴奋性反应的细胞倾向分布于具有Ih电流的锥体神经元中。
Objective To investigate the effect of orexinA on the PL cells of mouse prefrontal cortex by patch clamp whole cell recording. Methods The prefrontal cortical slices of mice were prepared and the electrophysiological characteristics of pyramidal neurons and interneurons were observed and recorded in patch-clamp whole-cell recording mode and their response to orex-inA. Results The total response rate of orexin A in 54 cases of pyramidal neurons was 51.9%. 36 cases of Ih (+) pyramidal neurons had a response rate of orexinA to orexinA of 66.7%, 18 cases of Ih (-) pyramidal neurons to orexinA rate of 22.2%, the difference was statistically significant (P <0.01); 15 Example interneurons did not respond to orexinA. CONCLUSION: orexinA has some excitatory effects on the pyramidal neurons in PL zone of prefrontal cortex, and the cells expressing the excitatory response tend to distribute in pyramidal neurons with Ih current.
