OBJECTIVE To investigate the effects of CIG,an ingredient extracted from a traditional Chinese herb Cornus officinalis,on neurological function and infarct size after ischemic stroke.METHODS SD rats underwent 90 min of middle cerebral artery occlusion(MCAO).CIG was intragastrically administered to rats in dose of 60 mg·kg-1·d-1,starting 6 h after the operation.The behavioral test was performed by using the modified neurological severity score(mNSS) at 2,7,14,21,28 d.Magnetic resonance imaging(MRI) was conducted at 2,7,14,and 28 d and then sacrificed and the infarct volume was measured using 2,3,5-Triphenyltetrazolium chloride(TTC) staining.RESULTS The treatment with CIG significantly improved neurological function at 7,14,and 28 d.MRI imaging demonstrated that total lesion volumes were reduced in the CIG group compared with the model rats at 7,14,28 d.TTC staining showed that infarct volumes of the CIG group were significantly reduced at 28 d.CONCLUSION CIG treatment after 6 h ischemia improved neurological function and reduced infarct size after ischemia in rats.
